I know this one is impossible for now given the state of html5 audio (unless I missed some important news), but I want to put it out here so it sits in the back burner while more important dishes are being cooked:
It'd be nice to be able to have detailed information about the audio currently being played (be it background music or sound effect).
How detailed? Enough as to be able to build mechanics similar to equalizer-graphs common in audio player software. This would enable developers to change the game's appearance based on currently played sounds, be it as simple as making a character's mouth move when it's voice track is producing sound (and leave it closed while it is playing but becomes silent), adding flashes to the background of a game according to some heavy music or even some full gameplay mechanics like in the game "Beat Hazard".
